PhD position at MycSA: ‘To decipher the modulation of the biosynthesis of enniatines by Fusarium spp. under the effect of abiotic stresses.’

The quality of cereals and derived products is significantly impacted by the contamination of the grains by mycotoxins. Among the most alarming mycotoxins are the toxins produced by the species of Fusarium. Some of these toxins were largely studied and are the object of a European regulation (regulation EC 1126/2007). Others, indicated under the term of emergent, because put in obviousness recently in harvests, are the object of discussion; a regulation fixing the values thresholds of contamination should be soon published. Enniatines are such emergent mycotoxins. Nowadays, fragmented knowledge on the mechanisms of accumulation of the enniatines in the grains limit the definition of strategies of control and does not allow to guarantee harvests with minimal levels of contamination. It is also impossible now to predict which will be the evolution of the risk related to the enniatines in a context of climate changes, which, as that was shown for other toxins, are likely significantly to impact the levels of contamination of harvests. Therefore, the objective of this project of doctorate is to progress in knowledge on the regulation of the biosynthesis of enniatines and its role in the adaptive response of the producing strains to the abiotic factors.

The project will be centered around the following direct question:  Up to what point and by which biological systems the production of mycotoxins of the enniatine type is under influence of environmental factors being able to be exacerbated by the climate changes? This question will be tackled through 3 items: (1) Which are the effects of temperature, water and oxidizing stresses on the production of enniatines? (2) Which are the mechanisms of concerned molecular regulation and which is the role of transcription factors ? (3) How do the strains producing enniatines behave within cocktails of Fusarium strains of  other toxinogenic species, when they are subjected to the abiotic stresses?

Know-how: the candidate will profit from the skills of the MycSA’s team for the study of the mechanisms of biosynthesis of secondary metabolites by filamentous mushrooms.

Competences acquired during the thesis: This project will make it possible to the candidate to acquire many technical and theoretical skills necessary to the comprehension of the mechanisms of regulation of the biosynthesis of fungal secondary metabolites: fungal cultures, analytical chromatography (LC and UPLC/DAD, LC/MS), fungal genetics (construction of mutants), molecular biology (QPCR and RT-QPCR).

Necessary field of competence to apply: Academic knowledge in biology, microbiology, and genetics of filamentous mushrooms. Competences in analytical techniques, control of the techniques of basic molecular biology (extraction DNA and ARN, PCR, RTPCR, electrophoresis).
